Projets récents

 

Les travaux de virologie moléculaire effectués dans mon laboratoire reposent sur l'étude de trois virus : parvovirus porcin (PPV), le virus de la diarrhée virale bovine (BVDV) et les densovirus (entomoparvovirus). Nous étudions, d'une part les mécanismes moléculaires impliqués dans l'infection et d'autre part, la relation structure-fonction des protéines de capside virales. Nous cherchons à déterminer la structure 3D des deux parvovirus ainsi que certaines protéines non-structurales. Nous travaillons également à déterminer la structure de la gp53 du BVDV afin de raffiner un nouveau type de vaccins.
 
 
Base moléculaire du tropisme du parvovirus porcin (PPV)
 
Le parvovirus porcin cause des problèmes reproducteurs chez le porc. Afin de mieux comprendre le rôle des acides aminés caractérisant les virus pathogènes, nos travaux ont pour objectif d'élucider quelles sont les interactions virus-cellules impliquant ceux-ci. Nos travaux récents ont démontré que les différences moléculaires qui caractérisent les souches de PPV sont uniquement localisées au sein des protéines structurales (Bergeron, Hébert et Tijssen, 1996). De plus, nous avons développé un système cellulaire in vitro permettant de distinguer la souche vaccinale des souches pathogéniques. Ce système nous permet ainsi d'étudier la permissivité des cellules pour des souches chimériques et/ou mutantes produites en laboratoire. C'est ainsi que nous avons démontré, par la construction de virus chimériques, qu'un maximum de trois acides aminés localisés dans VP1/2 sont nécessaires à la différence de permissivité observée.

Création d'un vaccin recombinant sous-unitaire composé de la gp53 du BVDV
 
Bien que le BVDV soit le pathogène principal chez les bovidés (pertes de 10M$ annuellement), les souches étaient néanmoins peu virulentes comparativement aux souches ayant fait apparition au Québec en 1993. Nous avons caractérisé ces souches et nous avons démontré que de nouveaux vaccins étaient requis. Nous travaillons présentement à la préparation d'un vaccin sous-unitaire dans différents systèmes d'expression (baculovirus, Pichia, Sindbis).

Organisation génomique et stratégie de transcription des densovirus
 
Les densovirus sont des parvovirus d'insectes très différents des autres parvovirus tant sur le point de vue de l'organisation génomique que de la stratégie de transcription. Nous sommes présentement à étudier la régulation de la transcription des gènes de la capside virale afin d'optimiser l'expression dans un système baculovirus. Les protéines ainsi produites s'auto-assemblent lors de l'expression et peuvent devenir un véhicule pour l'expression d'épitopes de virus pathogènes afin de créer un vaccin efficace et sans danger pour les animaux ou pour les humains.
